Novel monoclonal antibody inhibits tumor growth in breast cancer and angiosarcoma

Apr. 19, 2013 ? A monoclonal antibody targeting a protein known as SFPR2 has been shown by researchers at the University of North Carolina to inhibit tumor growth in pre-clinical models of breast cancer and angiosarcoma.

In a paper published in the April 19 issue of Molecular Cancer Therapeutics, a team led by Nancy Klauber-DeMore, MD, Professor of surgery and a member of UNC Lineberger Comprehensive Cancer Center, used a monoclonal antibody to target SFRP2 expressed in cells from triple-negative breast cancer and the aggressive blood-vessel malignancy angiosarcoma, reducing the rate of tumor growth. The antibody, created at the University of North Carolina, is the first therapeutic discovered that targets SFRP2.

"We showed in this paper that targeting SFRP2 with a monoclonal antibody in pre-clinical models inhibits tumor growth. This demonstrates that SFRP2 is a therapeutic target for cancer" said Dr. DeMore.

The DeMore lab first discovered the role of SFRP2 in tumor growth while looking to develop an alternative to the FDA-approved anti-angiogenesis drug known as Avastin (bevacizumab). Avastin targets the protein VEGF, which has also been tied to angiogenesis (the production of new blood vessels). Although Avastin is of benefit to some patients with cancer, not all tumors respond to Avastin, and of those that respond, some eventually progress. To find a solution for patients whose tumors are resistant to Avastin, DeMore began looking at other proteins linked to angiogenesis that could be used as therapeutic targets.

"We previously microdissected blood vessels from malignant human breast cancers and compared gene expression to blood vessels microdissected from normal tissue. We found a number of genes that were highly over-expressed in the malignant blood vessels compared to normal. One of those genes was SFRP2," said Dr. DeMore.

The DeMore lab found that SFRP2 is expressed in a variety of human cancers, including breast, prostate, lung, pancreas, ovarian, colon, kidney tumors, and angiosarcomas, DeMore, working with Dr. Cam Patterson, Ernest and Hazel Craige Distinguished Professor of Cardiovascular Medicine, discovered that SFRP2 acted as a potent stimulator of angiogenesis, leading their team to hypothesize that targeting SFRP2 could inhibit tumor growth. In collaboration with Dr. Russ Mumper, the John A. McNeill Distinguished Professor in the Division of Molecular Pharmaceutics, their group developed a drug to target SFRP2.

"Demonstrating that a monoclonal antibody to SFRP2 inhibits tumor growth in pre-clinical models opens up a new potential for drug development. This treatment is not presently available for human studies, but our efforts are focused on obtaining funding for further drug development that would lead to a clinical trial" said DeMore.

The vTrue strikes out only on a comfort level?these headphones are bulky, and after a while, you're likely to feel some pressure on your scalp.

On Bill Callahan's "Drover," the vTrue graces his vocals with a nice treble edge which helps it stand front and center in the mix. This is important, because the boosted bass response arms the constant drumbeat with some serious low-end thunder, making it also stand out quite a bit. Without the sculpted treble, his vocals would get lost and the mix would be too focused on the low-mids and lows.

Jay-Z and Kanye West's "No Church in the Wild" benefits from the high frequency sculpting, as well?the kick drum loop has a nice punchy attack, while the sub-bass synth hits that sit beneath the loop in the mix are delivered with some serious throttle. The vocals in this dense mix stand out in the same way Callahan's do on his track?the crisp edge they get from the vTrue helps separate them from the powerful lows and mids.

Classical tracks, like John Adams' "The Chairman Dances," can sound a tad too tweaked and sculpted. The highs are boosted, which occasionally makes the higher register strings and brass sound a bit too bright, but the lower register strings absorb the added bass response nicely. The large drum hits at the end of track sound a bit too bass-boosted, however?almost as if they are amplified instead occurring naturally in a concert hall.

In other words, these headphones don't offer a flat response in any sense?there's tons of bass, and lots of liberties are taken with the high-mids and highs, as well. The result is something bass lovers will enjoy and purists will probably want to avoid. If you're looking for headphones in the price range with a bit less boosting in the lows, the?Sennheiser Momentum?is a solid option and less bulky, while the TDK ST750 is a powerful pair that focuses more on the high-mids and highs.?If you like big bass, but the vTrue's price tag is too high, the?Denon Urban Raver AH-D320?offers a similar sound signature for a significantly lower price. And if it's a true studio headphone pair you seek, you can spend quite a bit less and still have top quality?Sennheiser's HD 280 Pro?is an excellent flat response option. The Velodyne True offers clean, bold performance with booming low-end, and it comes with two top-notch, detachable cables?if it were less expensive, it would get a higher rating.

Apr. 19, 2013 ? Scientists have revealed how a bacterial enzyme has evolved an energy-efficient method to move long distances along DNA. The findings, published in Science, present further insight into the coupling of chemical and mechanical energy by a class of enzymes called helicases, a widely-distributed group of proteins, which in human cells are implicated in some cancers.

The new helicase mechanism discovered in this study, led by researchers from the University of Bristol and the Technische Universit?t Dresden in Germany, may help resolve some of the unexplained roles for helicases in human biology, and in turn help researchers to develop future technological or medical applications.

A commonly held view of DNA helicases is that they move along DNA and "unzip" the double helix to produce single strands of DNA for repair or copying. This process requires mechanical work, so enzyme movement must be coupled to consumption of the chemical fuel ATP. These enzymes are thus often considered as molecular motors.

In the new work, Ralf Seidel and his team at the Technische Universit?t Dresden developed a microscope that can stretch single DNA molecules whilst at the same time observe the movement of single fluorescently-labelled helicases. In parallel, the Bristol researchers in the DNA-Protein Interactions Unit used millisecond-resolution fluorescence spectroscopy to reveal dynamic changes in protein conformation and the kinetics of ATP consumption.

The team studied a helicase found in bacteria that moves along viral (bacteriophage) DNA. The work demonstrated that, surprisingly, the enzyme only consumed ATP at the start of the reaction in order to change conformation. Thereafter long-range movement along the DNA was driven by thermal motion; in other words by collisions with the surrounding water molecules. This produces a characteristic one-dimensional "random walk" (see picture), where the protein is just as likely to move backwards as forwards.

Mark Szczelkun, Professor of Biochemistry from the University's School of Biochemistry and one of the senior authors of the study, said: "This enzyme uses the energy from ATP to force a change in protein conformation rather than to unwind DNA. The movement on DNA thereafter doesn't require an energy input from ATP. Although movement is random, it occurs very rapidly and the enzyme can cover long distances on DNA faster than many ATP-driven motors. This can be thought of as a more energy-efficient way to move along DNA and we suggest that this mechanism may be used in other genetic processes, such as DNA repair."

The work in Bristol has been funded by the Wellcome Trust through a programme grant to Professor Mark Szczelkun from the School of Biochemistry.

"These days you can safely glue on so you don't damage the jewelry, and then the girl has something to keep after prom instead of us being 20 years later with dry old roses. You've actually got this bracelet or this necklace that you can look back at and go, 'Oh, that was so much fun,'" said Della Mendenhall, a manager and product developer at Gillespie Florists in Indianapolis.

Boutonniere holders (think ice cream cone shape) often anchor the traditional male floral in metal. They come in filigree, vine and many other designs. Magnet sets can be used to keep them in place, and they can be reused for high schoolers who plan to attend more than one prom.

Sparkly broaches or decorative pins can also be used as an attachment for teens of any gender.

Embellishments

McCormick is a low-beta stock that has proven that it?s worth accumulating when it?s down.

The stock is pricey, compared to its peers in the processed and packaged goods subsector, especially given its modest earnings expectations.

There is growing concern about the outperformance of consumer staples and healthcare stocks. The Procter & Gamble Company (NYSE/PG) is up more than 10 points on the stock market since January.

This ?safe haven? breakout is still positive, but earnings have to deliver. All kinds of brand-name blue chips have run up tremendously?almost unbelievably.

But it will all be just a trade if earnings results don?t show growth. So far, numbers are coming in a little bit light. But business conditions in the first quarter were expected to be slow, just like gross domestic product (GDP) in the fourth quarter of 2012.

Investor sentiment still isn?t strong enough to carry this market higher on just consumer staples and healthcare stocks. Broadening is the key for the near-term rally to continue.
